Stem cells found in adults may repair nerves. Feb 2004 - A news release from the American Association for the Advancement of Science (AAAS) details current research findings on nerve regeneration. In a dramatic display of stem cells' potential for healing, a team of Johns Hopkins scientists reports that it has engineered new, completed, fully working motor neuron circuits — neurons stretching from spinal cord to target muscles — in paralyzed adult animals. The research, in which mouse embryonic stem cells were injected into rats whose virus-damaged spinal cords model nerve disease, shows that such cells can be made to retrace complex pathways of nerve development long shut off in adult mammals, the researchers say.

Bone marrow (BM) contains hematopoietic stem cells (HSCs), which differentiate into every type of mature blood cell; endothelial cell progenitors; and marrow stromal cells, also called mesenchymal stem cells (MSCs), which can differentiate into mature cells of multiple mesenchymal tissues including fat, bone, and cartilage. Recent findings indicate that adult BM also contains cells that can differentiate into additional mature, nonhematopoietic cells of multiple tissues including epithelial cells of the liver, kidney, lung, skin, gastrointestinal (GI) tract, and myocytes of heart and skeletal muscle (Blood. 2003 Nov 15;102(10):3483-93).

Human marrow stromal cells subjected to a simple treatment protocol induced bone marrow stromal cells to exhibit a neuronal phenotype, expressing neuron-specific proteins and to differentiated into neurons. Therefore, adult marrow stromal cells can be induced to overcome their mesenchymal commitment and may constitute an abundant and accessible cellular reservoir for the treatment of a variety of neurologic diseases. Future use of adult stem cells: In the future, it may be possible for a person to...

Future use of adult stem cells: In the future, it may be possible for a person to use a sample of his or her own stem cells to regenerate tissue, which would reduce or even eliminate the danger of rejection. How might this be done? Some possibilities include:

• Collecting healthy adult stem cells from a patient and manipulating them in the laboratory to create new tissue. The tissue would be re-transplanted back into the patient's body, where it would work to restore a lost function.
• Therapeutic cloning might enable the creation of embryonic stem cells that are genetically identical to the patient.
• One less invasive way to achieve this goal would be to manipulate existing stem cells within the body to perform therapeutic tasks. For example, scientists might design a drug that would direct a certain type of stem cell to restore a lost function inside the patient's body. This approach would eliminate the need for invasive surgical procedures to harvest and transplant stem cells.
